if you are doing this quest for 1st time you get the 1st(4 star quest)? and for next times you get the 2nd quest(5 star quest)??

Both gives the job permit. You can choose which one you prefer to do. There are no prequest for this.

Better to do the "The depths of the wines of the world" as all you have to do is sail and talk to a bunch of people. The other is a lot easier though if you have or can find 200 tequila's.
