Looks like this is basically for the Nanban skill although it could be an alternative to TLG as a merchange marine ship. Manilla Galleon is a bigger barge.

Some EA ships can be built in EA ports just like any other ships, all you need is EA port permit unlocked. I assume you're asking about this ship in particular, which needs to be built with Special Shipbuilding in Edo with r18 SB, using the hull shown above (E.Asian Heavy). Click the link to see where to make the hull. You'll need 3 of the uncommon & expensive East Asian Shipbuilding Techniques items.

To obtain east asian sb technique, you need to do nanban trade until you unlock the final town (where the historical npc resides): Chongqing (China), Anping (Taiwan), Edo (Japan), or Busan (Korea). Then fill your cargo with copper ore or some tradegood from nearby SEA, keep doing nanban trade (trade 50x copper ore at a time->port reset->trade->reset...) in one of the aforementioned towns, and there's a 10% chance of you being summoned by the npc for a random reward. 5x east asian sb techniques is one of the rewards.
